چکیده

A large number of human genes have been found to contain more than one polyadenylation site (poly(A) site). These lead to multiple messenger ribonucleic acid (mRNA) transcripts, although some of them do not yield protein products. Like alternative initiation and alternative splicing, alternative polyadenylation (APA) contributes to the complexity of the overall pool of mRNA transcripts in human cells. However, little is known about how the cell utilizes different poly(A) sites in response to different cell conditions and developmental stages. Several protein factors have been identified as being necessary for in vitro cleavage and polyadenylation. If the gene of a polyadenylation factor has multiple poly(A) sites, producing variable protein products with different functions, then their APA can control the polyadenylation activity under different cell conditions. The dynamics of the gene expression levels among APA products so far has been studied by experimental approaches only. Here we propose a novel mathematical model consisting of differential equations with a time delay to study the dynamical behavior of the gene expression levels. Numerical simulations have been provided for some of the theoretical analyses of the equations. Our model suggests an oscillatory pattern of APA of CSTF3, indicating APA is employed to maintain homeostasis of its function, perhaps general polyadenylation activity as well, in the cell.

I-13 Infertility with Impaired Zona Pellucida Adhesion of Spermatozoa from Mice LackingTauCstF-64

Background: Fertilization is a multistep process requiring spermatozoa with unique cellular structures and numerous germ cell-specific molecules that function in the various steps. In the highly coordinated process of male germ cell development, RNA splicing and polyadenylation help regulate gene expression to ensure formation of functional spermatozoa. Male germ cells express tauCstF-64 (Cstf2...

P-215: Discovery of A Novel APA Variant of A Human Potential Gene Based on Expressed Sequenced Tags Analysis

Background: Expressed sequence tags (ESTs) are sequences of cDNA fragments prepared from different tissue sources. There are over one million of these sequences in the publicly available database, and these sequences are believed to represent more than half of all human genes. The ESTs belong to different cDNA libraries, was prepared from one particular cell type, organ, or tumor. Therefore, th...

Alternative polyadenylation sites reveal distinct chromatin accessibility and histone modification in human cell lines

MOTIVATION In addition to alternative splicing, alternative polyadenylation has also been identified as a critical and prevalent regulatory mechanism in human gene expression. However, the mechanism of alternative polyadenylation selection and the involved factors is still largely unknown. RESULTS We use the ENCODE data to scan DNA functional elements, including chromatin accessibility and hi...

A large-scale analysis of mRNA polyadenylation of human and mouse genes

mRNA polyadenylation is a critical cellular process in eukaryotes. It involves 3' end cleavage of nascent mRNAs and addition of the poly(A) tail, which plays important roles in many aspects of the cellular metabolism of mRNA. The process is controlled by various cis-acting elements surrounding the cleavage site, and their binding factors. In this study, we surveyed genome regions containing cle...

Alternative polyadenylation of cyclooxygenase-2

A biologically important human gene, cyclooxygenase-2 (COX-2), has been proposed to be regulated at many levels. While COX-1 is constitutively expressed in cells, COX-2 is inducible and is upregulated in response to many signals. Since increased transcriptional activity accounts for only part of the upregulation of COX-2, we chose to explore other RNA processing mechanisms in the regulation of ...
